ISSN 0333-1024There is now evidence that melatonin may have a role in the biological regulation of circadian rhythms, sleep, mood, and ageing. Altered melatonin levels in cluster headache and migraine have been documented. Melatonin mechanisms are related to headache pathophysiology in many ways, including its anti-inflammatory effect, toxic free radical scavenging, reduction of proinflammatory cytokine up-regulation, nitric oxide synthase activity and dopamine release inhibition, membrane stabilization, GABA and opioid analgesia potentiation, glutamate neurotoxicity protection, neurovascular regulation, serotonin modulation, and the similarity of chemical structure to that of indomethacin. Treatment of headache disorders with melatonin and other chronobiotic agents is promising. A double-blind, placebo-controlled trial shows melatonin is effective in cluster headache prevention, other studies also show benefit in other disorders. Melatonin plays an important role in headache disorders, offering new avenues for studying their pathophysiology and treatment. PMID: 15910564

| Proof of how slowly these things can progress.... "Melatonin And Cluster Headaches" In this study, 10 mg of melatonin was administered to half of 20 cluster-headache patients in a double-blind controlled study that lasted 14 days. Headache frequency was significantly reduced in the melatonin treatment group. Five of 10 people in the melatonin group reported that their attack frequency declined after only 30-35 days of treatment. No patient in the placebo group responded." Bob, Do you know if this was quoting the study from '96 or was this based upon a new study of melatonin and clusters? thanks, Bobw

I just checked it out. It's the same old '96 paper. It also mentions this one where melatonin helps two people with chronic refractive CH: Peres MF, Rozen TD. Melatonin in the preventive treatment of chronic cluster headache. Cephalalgia 2001; 21:993–5. Jesse

Yay for melatonin!! There are 28 citations on Pubmed that include both "cluster headache" and "melatonin" - though not all are studies, most are lists of therapies that have been used.

I tried it this cluster. 9mg / night for 5 or 6 weeks. Didn't do anything (but I may have done it wrong). I quit taking it this past weekend and I think I'm sleeping better than I did while taking it.

Bob, Bob and Bob......... Bob (not Bob or Bob) screwed up. He only took 9 mg instead of the directed 10. I don't know whether to listen to Bob, Bob, or Bob. Melatonin did nuthing for me, unless it made me sleep like crap. Like Bob, but not Bob or Bob. I'm losing my mind..... RJ

I'm thinking that since melatonin is an indole that the shrooms probably interfered with the melatonin. Seriously, even before I was in cycle I would take mel before bed occasionally. 1.5mg would help me sleep. 3mg would interfere with sleep, shorter an more restless sleep. 9mg didn't seem to do anything. Can the cluster brain be that messed up?

Bob P has it right. A good study reported on Medscape showed that, for sleep, a dose of 0.3mg was best. Using more or less--for sleep--was less effective.
